Callus induction and production of total saponins in Pfaffia glomerata (Spreng.) Pedersen in vitro

There was the influence of various media constituents on the growth and saponin production in Pfaffia glomerata callus. Chemical analysis of the saponin content in callus culture from shoot and root explants of P. glomerata, were carried out in order to determine whether it was possible to obtain secondary metabolites in vitro. The effects of auxin (2,4 D) and cytokinins (KIN) on saponin content and callus growth were examined; a better increased amounts of saponins was observed when combining 1,0 mg.L⁻¹ 2,4-D and 0.5 mg.L⁻¹ KIN (shoot explant: 4,09% and root explant: 3,81%). The total saponins content of P. glomerata roots and shoots in vivo was 3,74% and 2,69% respectively. High sucrose concentrations (60 g.L⁻¹) increased the content of total saponins (5,92%).
